POP1 is a component of Ribonuclease P that cleaves the 5' leader from precursor tRNAs. The C-terminal domain of POP1 interacts with H1 RNA and has a stabilizing role. This domain adopts a beta-barrel structure that is structurally very similar to the C-terminal beta-barrel domains of eIF2 gamma, EF-Tu and tRNA thiouridylase MnmA [1].

These RNA-containing enzymes generate mature tRNA molecules by cleaving their 5' ends. This is a short N-terminal domain consisting of a small helical core and a long alpha-helix [2], which is enriched with positively charged residues involved in pre-tRNA substrate recognition [2].

The archaeal proteins are found to be expressed within ribosomal operons and several of the sequences are described as ribonuclease P protein subunit p29 proteins [1,2]. The structure of the RNase P subunit, Rpp29, from Methanobacterium thermoautotrophicum has been determined. Mth Rpp29 is a member of the oligonucleotide/oligosaccharide binding fold family. It contains a structured beta-barrel core and unstructured N- and C-terminal extensions bearing several highly conserved amino acid residues that could be involved in RNA contacts in the protein-RNA complex [2]. Rpp29 catalyses the endonucleolytic cleavage of RNA, removing 5'-extranucleotides from tRNA precursor. It interacts with the Rpp25 and Pop5 subunits.

Downstream of Notch may repress the expression of proneural genes and inhibit neuronal differentiation thereby maintaining neural progenitors [1]. NEPRO may also play a role in preimplentation embryo development [2]. NEPRO interacts with multiple protein subunits of RMRP and is implicated in Anauxetic dysplasia 3 [3]. NEPRO is likely the human orthologue of RMP1 subunit from yeast and they share a four-helix bundle domain structure.
