This domain consists of the amino-terminal half of the Klenow fragment in E. coli polI it is also found in the Werner syndrome helicase (WRN), focus forming activity 1 protein (FFA-1) and ribonuclease D (RNase D). Werner syndrome is a human genetic disorder causing premature aging; the WRN protein has helicase activity in the 3'-5' direction [4,5]. The FFA-1 protein is required for formation of a replication foci and also has helicase activity; it is a homologue of the WRN protein [3]. RNase D is a 3'-5' exonuclease involved in tRNA processing. Also found in this family is the autoantigen PM/Scl thought to be involved in polymyositis-scleroderma overlap syndrome.

The Egalitarian (Egl) protein functions as part of a complex with BicaudalD (BicD) and dynein light chain (DLC-1) to recruit the dynein motor complex to specific cargoes and regulate dynein activity. In C. elegans, the EGAL-1 protein (homolog of Egalitarian) interacts with both the BICD-1 and DLC-1 proteins. Genetic disruption of egal-1 leads to defects in hyp7 nuclear migration, similar to defects seen when disrupting bicd-1 or components of dynein. The egal-1 phenotype is enhanced when combined with a nud-2 mutant, but not enhanced by bicd-1, suggesting egal-1 and bicd-1 function in the same genetic pathway. A model is proposed where BICD-1, EGAL-1 and DLC-1 form a complex that is recruited by the nuclear envelope protein UNC-83. This complex then helps recruit the dynein motor to the nuclear envelope to regulate nuclear migration [1-2].
